Día antes: Coating ELISA NF155, CASPR1, CNTN1, NF140

[CASPR1]i = 0,789 mg/ml, [NF155]i = 0,20 mg/ml, [CNTN1]i = 0,25 mg/ml,[NF140]i = 0,25 mg/ml

Buffer coating: carbonate-bicarbonate (100 ml agua destilada + 1 pastilla C3041 Sigma)

  • CASPR1: 48 pozos []f = 5 ug/ml: 2,5 ml buffer +   15,8 ul
  • NF155: 48 pozos []f = 1 ug/ml: 2,5 ml buffer +   12,5 ul
  • CNTN1: 48 pozos  []f = 1 ug/ml: 2,5 ml buffer +   10 ul
  • NF140: 48 pozos []f = 5 ug/ml: 2,5 ml buffer + 50 ul

Protocolo:

  • Bloquear con leche 5% en PBS-tween 0’1% à 200 ul pozo (incubar 1h a temp. ambiente)
  • Lavar 3 veces con PBS-tween 0’1%
  • Diluir los  sueros:
    • Screening:  diluir sueros 1/100 en leche 5% en PBS-tween 0’1%
  • Incubar con los sueros 1h
  • Lavar 3 veces con PBS-tween 0’1%
  • Incubar 45min con RAH IgG HRP 1/3000 en leche 5% en PBS-tween 0’1% à 100 ul/pozo
  • Lavar 3 veces con PBS-tween 0’1%
  • Incubar 5min con TMB: preparar solución un rato antes y tapar de la luz (para que se atempere) à 100 ul/pozo
  • Parar la reacción con 50ul de H2SO4 25%
  • Leer a 450-620 nm
