20180926: DRG neurons ICC (Zika willison + unit patients + controls)

Samples:

• IgG:
  • unit patients
  • controls (healthy, ELA and dysferlin patients)
• IgM:
  • unit patients
  • Zika (Willison) 
  • controls (healthy, ELA and dysferlin patients)

DRG NEURONS ICC

• Incubate with patient’s sera in a 24 wells plate: 1 hour 37ºC  (prepare the dilution in the wells and then put one coverslip per well)
  • For IgG: 2 µl of sera in 300 µl of NG medium
    
  • For IgM: 7,5 µl of sera in 300 µl of NG medium
• Fix with PFA 4 % –> 10 min at room temperature
• Wash 1 time with PBS1x
• Incubate with secondary antibody (1 hour at RT) –> GAH 488 IGg/IgM 1:1500 diluted in Goat serum 5%
• Wash 1 time with PBS1x
• Mount with Vectashield
