Objectives: To test patient 18-532 for IgM positivity towards NF140.
Patient: 18-352
NF140 ICC:
- HEKs 293 Cells: 2017.06.27
- Blocking solution: 5% goat serum in PBS.
- Patient’s sera diluted 1/40 and 1/10 for IgM with 5% goat serum in PBS.
- Primary antibody: AF3235 diluted 1/1000.
- Secondary antibodies: GAH 594 IgM (1/1000) + GAC 488 (1/1000).
- Vectashield mounting medium
Results: We saw a positive result in the sample diluted at 1/10 and a mild positive result in the sample diluted at 1/40, very similar to the positive result in non-transfected cells diluted at 1/40 (experiment on 2018.01.25). We are going to repeat the experiment in non transfected cells with the same sera diluted at 1/10 and we are going to perform a teased nerve fibers immunohistochemistry.
