Protocolo:

  • Lavar 3 veces con PBS-tween 0.1%
  • Bloquear con leche 5% en PBS-tween 0’1% – 200 ul pozo (incubar 1h a temp. ambiente).
  • Suero 1/100 en leche
  • Incubar con los sueros 1h
  • Lavar 3 veces con PBS-tween 0’1%
  • Incubar 45min con anticuerpo secundario:
    • RAH IgG HRP 1/3000 en leche 5% en PBS- tween 0’1% à 100 ul/pozo
    • IgG1, IgG2, IgG3, IgG4 1/500 en leche 5% en PBS-tween 0’1% – 100 ul/pozo
  • Lavar 3 veces con PBS-tween 0’1%
  • Incubar 5min con TMB: preparar solución un rato antes y tapar de la luz (para que se atempere) à 100 ul/pozo
  • Parar la reacción con 50ul de H2SO4 25%
  • Leer a 450/570
