20190918: IHQ – Peripheral nerve monkey Euroimmun (prueba)

Patients:

5. Commercial Caspr1 mAb (rabbit) 1:25

IHC: 

• Comercial tissue sample: FA 1120-1005 (Euroimmun)
• Incubate with blocking solution goat 5% in PBS for 30 min
• Patient’s sera diluted 1/10 in PBS-tween –> put 30 microl of the dilution in every well of the reactives support. Then, put the slide on.
• Wash the slide 2 times with PBS-tween (with shaking)
• Put 25 microl of the secondary antibody (GAH 488 monkey abs 1:200; GAR 488 1:250) in every well of the reactives support. Then, put the slide on.
• Wash the slide 2 times with PBS-tween (with shaking)
• Mounting medium from the kit

Results: there are no differences between the different samples, there is an unspecific stainning with strong background and I don’t see paranodes (only with Caspr1 mAb that is completely negative)
