Objective: To test IgG towards ANO2 in MS patients and to check if the sample 153 is positive.
Samples: 153 (serum), 328 (LCR), negative control (serum)
ICC ANO2 – CONDITION 1 (with primary antibody):
- Blocking solution: Goat serum 5% in PBS
- Patient’s sera diluted 1/100 (LCR diluted 1/40)
- Primary antibody: ANO2 TA 316534 Origene diluted 1/500 in Goat 5%
- Secondary antibodies: GAR488 + GAH 594 Jackson diluted 1/1000
- Vectashield mounting medium
ICC ANO2 – CONDITION 2 (without primary antibody)
- Blocking solution: Goat serum 5% in PBS
- Patient’s sera diluted 1/100 (LCR diluted 1/40)
- Secondary antibodies: GAH 594 Jackson diluted 1/1000
- Vectashield mounting medium
RESULT: all the samples are negative!!!!! We don’t have any positive patient against ANO2
