Objectives: To test Multiple Sclerosis patients for IgG positivity towards ANO2
Patients: 124 to 145 (from the Multiple Sclerosis sera collection) + negative control
ANO2 ICC:
- HEKs 293 Cells Transfection: 12.03.2018
- Blocking solution: Goat Serum diluted 5% in PBS.
- Patient’s sera diluted 1/100 with 5 % Goat Serum
- Primary antibody: ANO2 (TA316534 OriGene) diluted 1/500.
- Secondary antibodies: GAR 488 + GAH 594 IgG (1/1000)
- Vectashield mounting medium.
Resultado: todas las muestras son negativas menos la número 145 –> sólo se ven positivas las células que son más positivas con el comercial de ANO2. Repetir esta muestra sólo con suero y con Ac secundario GAH594. También probar con cubres sin permeabilizar.
