Objectives
To test paraneoplastic neuropathies patients IgM staining pattern in order to dilucidate whether the staining seen in experiment 20170623 is just unspecific background staining. To do so, we will use CNTN2 transfected HEKs and as in the 20170623 experiment we will not use a commercial antibody.

CNTN2 ICC
- Human transfected CNTN-2 HEKs 293 grown in coverslips . Fixed with PFA 4% and blocked with 5% goat serum in PBS.
- PBS
- Blocking: 5% goat serum in PBS
- Patient’s sera diluted 1/40 (IgM) with 5% goat serum in PBS.
- Secondary Abs: GAH 594 (1/500) IgM
- Vectashield mounting medium
RESULTS
El marcaje apreciado en el experimento de hoy es el mismo que el visto en la ICC de Piezo 1 por IgM realizada el 23/06/2017. Como consecuencia, se concluye que dicho marcaje se corresponde al propio de las IgM, sin tener ninguna especificidad con el antígeno transfectado en las células HEK.
