Protocolo:
- Fijar 20 min con PFA 4%
- 1 lavado con PBS1x
- Bloquear con Goat serum 5% y 0’3% tritón (en PBS): 1h a RT
- Incubar con anticuerpos primarios diluídos en Goat serum 5% y 0’3% tritón (en PBS):
- 1 y 2) MBP y NF200
- anti-Myelin basic protein SMI99, 808401 (Biolegend). Dil. 1/60 (mouse): overnight 4ºC
- anti-Neurofilament200, N4142 (Merck). Dil. 1/60 (rabbit): 2 h RT
- 3 y 4) NF200 y MBP
- anti-Neurofilament200, N4142 (Merck). Dil. 1/60 (rabbit): overnight 4ºC
- anti-Myelin basic protein SMI99, 808401 (Biolegend). Dil. 1/60 (mouse): 2h a RT
- 5) suero CNTN1+: dil 1/75: overnight 4ºC
- 6) suero LIF+: dil 1/75: overnight 4ºC
- 7 y 8) suero Caspr1+: dil 1/75: overnight 4ºC
- 1 y 2) MBP y NF200
- 3 lavados con PBS1x
- Día siguiente: Incubar 1h con Ac. secundarios diluídos en Goat serum 5% y 0’3% tritón (todos a 1/500)
- 1) GAM488 (MBP) + GAR594 (NF200)
- 2) GAM594 (MBP) + GAR488 (NF200)
- 3) GAM488 (MBP) + GAR594 (NF200)
- 4) GAM594 (MBP) + GAR488 (NF200)
- 5-8) GAH 488 IgG
- 3 lavados con PBS1x
- Montar con Fluoromount
Resultado: no veo nada interesante… no se ve mielina
