Objective: To investigate Monkey’s Peripheral nerve IHC for IgG and IgM staining paterns of CIDP patients with NCAM Ab, a colocalization with NCAM commercial mAb.
Samples:
- For Ig G: 0103-00902 (27), 0103-00905 (28), 0302-00894 (29), 0701-00893 (30), 0702-00867 (31), 0702-00872 (32), 0702-00906 (33), 0802-00903 (34).
- For Ig M: 0907-00871 (25), 0103-00902 (27), 0103-00905 (28), 0302-00894 (29), 0701-00893 (30).
IHQ protolocol:
- We used commercial slides from Inova (peripheral nerve)
- Blocking with Goat Serum 5% in PBS (blocking solution). Incubate 1 hour at RT in a humid chamber.
- Incubate 35ul of sample diluated 1/20 (IgG) or 1/10 (IgM) in blocking solution 1h.
- Wash 2-3 times with PBS 1x.
- Incubate with anti-N-CAM commercial mAb (1/50) 1h.
- Wash 2-3 times with PBS 1x.
- Secondary Ab GAH monkey absorbed 488 and GAM594 (1/500) for 45min hour at RT.
- Wash 3 times with PBS 1x.
- Mount with Fluoromount mounting medium.
