Objective: To investigate Monkey’s Peripheral nerve IHC for IgG staining paterns healthy patients with NCAM Ab, a colocalization with NCAM commercial mAb. It is used as a control for the experiments of Amyotrofic Neuralgia.
Samples: 198-05 (05), 209-10 (10), 204-09 (9), 204-06 (6), 200-01 (1), 204-04 (4), 198-06 (6*), 202-03 (3), 206-01 (1), 204-16 (16), 204-18 (18), 151-07 (7), 192-02 (2), 207-04 (4), 204-12 (12).
IHQ protolocol:
- We used commercial slides from Inova (peripheral nerve)
- Blocking with Goat Serum 5% in PBS (blocking solution). Incubate 1 hour at RT in a humid chamber.
- Incubate 35ul of sample diluated 1/20 (IgG) or 1/10 (IgM) in blocking solution 1h.
- Wash 2-3 times with PBS 1x.
- Incubate with anti-N-CAM commercial mAb (1/50) 1h.
- Wash 2-3 times with PBS 1x.
- Secondary Ab GAH monkey absorbed 488 and GAM594 (1/500) for 1 hour at RT.
- Wash 3 times with PBS 1x.
- Mount with Fluoromount mounting medium.
