20210408: Monkey’s peripheral nerve IHC in healthy patients

Objective: To investigate Monkey’s Peripheral nerve IHC for IgG staining paterns of healthy patients with NCAM Ab, a colocalization with NCAM commercial mAb. It is used as a control for the experiments of Amyotrofic Neuralgia.

Samples:

  • For IgM: 198-05 (5), 209-10 (10).
  • For IgG: 204-11 (11), 202-16 (16), 195-12 (12), 204-07 (7), 220-24 (24), 207-03 (13), 198-01 (1), 206-02 (2), 198-03 (3), 204-19 (19) .

IHQ protolocol:               

  • We used commercial slides from Inova (peripheral nerve)
  • Blocking with Goat Serum 5% in PBS (blocking solution). Incubate 1 hour at RT in a humid chamber.
  • Incubate 35ul of sample diluated 1/20 (IgG) or 1/10 (IgM) in blocking solution 1h.
  • Wash 2-3 times with PBS 1x.
  • Incubate with anti-N-CAM commercial mAb (1/50) 1h.
  • Wash 2-3 times with PBS 1x.
  • Secondary Ab GAH monkey absorbed 488 and GAM594 (1/500) for 1 hour at RT.
  • Wash 3 times with PBS 1x.
  • Mount with Fluoromount mounting medium.
