20210406: Monkey’s peripheral nerve IHC in CIDP patients

Objective: To investigate Monkey’s Peripheral nerve IHC for IgG and IgM staining paterns of CIDP patients with NCAM Ab, a colocalization with NCAM commercial mAb.

Samples:

  • For Ig G: 0102-00861, 0401-00869, 0902-00879, 0902-00881, 0903-00856, 0903-00880, 0903-00889, 0907-00860, 0907-00871, 0912-00898.
  • For Ig M: 0503-00908, 0902-00881, 0903-00856, 0903-00880, 0903-00889.

IHQ protolocol:               

  • We used commercial slides from Inova (peripheral nerve)
  • Blocking with Goat Serum 5% in PBS (blocking solution). Incubate 1 hour at RT in a humid chamber.
  • Incubate 35ul of sample diluated 1/20 (IgG) or 1/10 (IgM) in blocking solution 1h.
  • Wash 2-3 times with PBS 1x.
  • Incubate with anti-N-CAM commercial mAb (1/50) 1h.
  • Wash 2-3 times with PBS 1x.
  • Secondary Ab GAH monkey absorbed 488 and GAM594 (1/500) for 45min hour at RT.
  • Wash 3 times with PBS 1x.
  • Mount with Fluoromount mounting medium.
