Objective: To test reactivity against Purkinje cells in MS patients and healthy controls (as controls in the previous NF155 IHC experiment)
Samples: MS patients (14 samples; 101-114; 2020.12.09) and HC (19 samples; 2020.12.16).
PROCEDURE:
- Incubate with blocking solution (goat serum 5% in PBS) 1h at RT.
- Incubate 50 microl of sample diluted 1/20 in goat serum 5% in PBS 1h at RT.
- Wash 3 times with PBS 1x.
- Secondary Ab GAH monkey absorbed (1/500) 1 hour at RT.
- Wash 3 times with PBS 1x.
- Mount with Fluoromount mounting medium.
RESULTS: MS samples and healthy controls have a very similar staining in Purkinje cells than NF155 patients, so it seems that the staining is unespecific (except for NF155 patients 1, 16 and 26). We are going to check it with mouse and mice cerebellum without fixation to see if the pattern is the same or not.
