20191030: Monkey’s peripheral nerve IHC in CIDP patients (CIDP samples from Holland and Birmingham)

Samples and protocol:

1. Universal blocking solution (1h) –> NCAM (mouse) 1:40 (1h) –> CNTN1 (goat) 1:50 (1h) –> 2º Ab: RAM488 (A11059) 1:400 + DAG555 1:400 (1h)
2. 19219 + paneurofascin mAb 1:50
3. 19200 + paneurofascin mAb 1:50
4. 19203 + paneurofascin mAb 1:50
5. Negative control

2-5: blocking goat 5% –> serum 1:20 (1h) –> 2º Ab GAC 594 1:500 + GAH 488 monkey absorbed 1:500

Results:

1. NCAM + CNTN1 –> colocalization ++ (NCAM = unmyelinated fibres, CNTN1 unmyelinated fibres and paranodes)
2. 19219 + paneurofascin mAb 1:50 –> intersticio + C schwann ?
3. 19200 + paneurofascin mAb 1:50 –> unspecific staining
4. 19203 + paneurofascin mAb 1:50 –> similar to NCAM, we will do doble staining
5. Negative control –> negative
