Objective: To investigate Monkey’s Peripheral nerve IHC staining paterns of CIDP patients with CNTN1 and CNTN1/Caspr1 Ab, a colocalization with CNTN1 commercial mAb. We are interesting in the staining of patched areas that could correspond to unmyelinated fibers
Samples:
- negative control 1:20
- Piffort 1:20
- Regine 1:20
- 16047 1:20
- CNTN1+ (15198) 1:20
Protocol: HRP + HEMATOXYLIN
- Incubate in hydrogen peroxide 0.3% x 15 min
- wash with PBS 1x
- Block with goat serum 5% in PBS 1h
- Incubate samples 1:20 for 2 hours
- Incubate with secondary Ab GAH IgH HRP (sigma A8419) 1:50 1 hour
- Wash with PBS
- Incubate with DAB (5 min)
- Wash with dH20
- Incubate with hematoxylin 30 seconds (not dilluted)
- was with dH20
- mount with DPX
Results:
It did not wor well. hematoxylin staining hides serum staining
