CNTN1 and CNTN1/Caspr1:
- Blocking solution: rabbit serum 1:40 in PBS
- Patients’ sera diluted 1:100 (rabbit serum 1:40 in PBS)
- Ac anti-CNTN1 diluted 1/1000
- RAH 594 IgG and RAG 488 IgG diluted 1/1000
- Vectashield mounting medium
Caspr 1:
- Blocking solution: goat 5% in PBS
- Patients’ sera diluted 1:100 ( goat 5% in PBS)
- Ac anti-Caspr1 diluted 1/250
- GAH 594 IgG and GAR 488 IgG diluted 1/1000
- Vectashield mounting medium
RESULTS:
Patients’ sera ( MRVM 2011, 17595 ) were positive for CNTN1 / Caspr1 co-transfection and colocalized with the commercial CNTN1 antibody
Sera from 15198 was positive when CNTN1 was transfected alone and
CNTN1 / Caspr1 were co-transfected
They also seemed positive when Caspr1 was transfected alone but did not colocalize with the Caspr1 commercial antibody. When CNTN1 was transfected alone, positivity was not clear.
