20190531: ICC in non-transfected cells and LRP4

Objective: To test possible unspecific staining in samples 18-860 (CIDP) and 101 (CMT).

Both samples are positive for NF155 (but no clear colocalization), positive ELISA 1/900, IH negative.

Samples: 18-860, 101, C+ (LAC)

ICC in non-transfected cells:

  • Blocking solution: Goat 5% in PBS (previously blocked with rabbit).
  • Primary antibody: AF3235 diluted 1/1000.
  • Secondary antibodies: GAC488 + GAH 594 IgG (1/1000).
  • Vectashield mounting medium.

RESULTS: Sample 18-860 has a mild unspecific staining in non-transfected cells, only with the primary antibody. Sample 101 is positive in LRP4-transfected cells, indicating an unspecific staining.
