20181003: ICC NF155 and CNTN1 in CMT patients

Objective: Detection of IgM or IgG positivity towards NF155 and CNTN1 of CMT and CIDP patients.

SAMPLES:

  • For NF155: IgG – 23 and 24; IgM – 53
  • For CNTN1: IgG: 12,24; IgM:18

NF155 ICC:

  • Blocking solution: 5% goat serum in PBS.
  • Patient’s sera diluted 1/100 for IgG and 1/40 for IgM with 5% goat serum in PBS.
  • Primary antibody: AF3235 diluted 1/1000.
  • Secondary antibodies: For IgG: GAC 488 + GAH 594 IgG (1/1000). For IgM: GAC488 + GAH 594 IgM (1/1000).
  • Vectashield mounting medium.

CNTN1 ICC: 

  • Blocking solution: Rabbit serum diluted 1/40 in PBS.
  • Patient’s sera diluted 1/100 for IgG and 1/40 for IgM with rabbit serum 1/40.
  • Primary antibody: AF904 diluted 1/1000.
  • Secondary antibodies: RAG 488 + RAH 594 IgG or RAH 594 IgM (1/1000)
  • Vectashield mounting medium

RESULTS: All samples were negative exept sample 23 for IgG towards NF155 and 24 for IgG towards NF155 (in sample 24 without a clear colocalization). We are going to perform an ELISA with NF155 protein.
