Objective
To verify that what we saw are nodes, we will mark this rat skin cuts with CASPR and neurofascina. We aim to see colocalizing positive marks.
Materials
see rat skin IHC 3
Procedure
see rat skin IHC 4
- Primary antibody: mouse-anti-CASPR 1:1000 + chicken-anti-neurofascin 1:500
- Secondary antibody: GAM- 594 1:500 + GAC-488 1:500
Results & conclusions
We can see some positive marks where colocalize CASPR and NFASC, so we conclude that we can see nodes in rat skin but a large time is needed to find them.
Photo 1. We can see a positive mark for NFASC155 that looks like a node.
Photo 2. We can see several positive marks for CASPR and one looks like a node. We see a lot of background because secondary antibody is antimouse (evolutionary close to rat).
Photo 3. We can see that the previous positive marks for NFASC155 and CASPR colocalize so there is a node of Ranvier.
